While hunting for food one day, Neville and Sam come into contact with a group of “Darkseekers” and nearly lose their lives in a petrifying incident. After almost dying, the officer comes up with the idea to capture one of the infected to test his cures on. To set the trap, Neville laid a tarp out that was rigged to collapse when triggered. Since the “Darkseekers” are attracted to blood, he broke a vile of his own blood on the tarp to draw them in. After capturing a female victim, he takes her to his lab for testing; this attempt at curing the disease seemed hopeless.
The following day, Neville noticed one of his mannequins in the city had been moved. Filled with confusion and frustration, he unloads a good amount of rounds into the mannequin that he called “Fred”. Neville went to investigate the scene and got caught in a snare trap waiting by what used to be a dummy. After waking up from unconsciousness, he realized that the sun was setting and that there wasn’t very much time left before he had to be back inside where it was safe.
To his surprise, the alpha male emerged with two infected canines by his side from the darkness of a building close to where Neville and Sam were. The dogs were released and sent to attack Neville and Sam, and during all of the chaos Sam was bit while defending his master. Neville was determined to save his best friend. With hopes high, Neville injected Sam with a strain of his serum. It was too late. By that time, Sam had started to show signs of infection, such as hair loss, discolored gums, and bloodshot eyes.
After Sam tried to attack Neville, the officer was forced to put his noble companion down by asphyxiation. Emotionally distraught, Neville couldn’t take it anymore. The next night, Neville decided to try and get back at the infected population for taking his best friend from him. He loaded up his SUV and went to a loading dock on the river where he planned to meet the insalubrious bunch. After killing many of them, they soon overcame his car and almost killed him, but just in the nick of time, a woman named Anna (Alice Braga) and a boy named Ethan (Charlie Tahan) slipped in and saved the almost dead man.
When they get back to Neville’s home, Anna is not aware of the precautions taken to stop the “Darkseekers” from following their trail, and that night they are tracked down and attacked. Preset UV lights were able to stop the first wave, Claymore mines were able to stop the second, but the third wave made it to the house with ease. In a panic, they try and hide to avoid being infected, but their attempts are not promising. They are forced into the basement(Neville’s lab) and are essentially cornered.

The lab is quickly flooding with the infected, and the three healthy people(Neville, Anna, and Ethan) are isolated in a glass room connected to the main part of the basement. The alpha male storms into the basement with rage and is determined to take down Neville. In an attempt to break the glass, the alpha male slams himself into it, but it’s not breaking, only cracking. These cracks begin to form a butterfly, so Neville remembers his daughter, who used to make a butterfly with her hands. Neville knows what he has to do.
With limited hope, Neville sent Anna and Ethan, with the cure in their possession, down a hidden passageway that lead to a safer place. Neville slowly reached into a drawer next to him and pulled out a hand grenade. He pulled the pin, and with full force, the alpha male and Neville ran at each other only to collide right as the grenade exploded. Anna and Ethan eventually made it to the survivors colony that had only been heard of. They delivered the cure upon their arrival, and Lieutenant Colonel Robert Neville’s legacy lived on.
